A Particulate Component Found in Nucleoli of Allium cepa and Vicia faba
نویسنده
چکیده
During an electron microscopic investigation of mitosis in the meristematic cells of Allium cepa and Vida faba, a particulate component of distinctive character has been observed in many nudeoli. The most striking feature of these granules in thin sections of osmium-fixed material (1) is their great electron density which makes them stand out dearly from the rest of the nudeolar material (Fig. 1). The majority of such particles are donghnut-like in appearance but, within one field, wide variations in shape are found. They may be round, oval, or rod-like in profile, possibly as a result of the angle of sectioning. In diameter they vary from approTimately 70 A to 200 A, with the majority at about 140 A (Figs. 2 and 3). Wide variations have also been observed in the occurrence of the particles. They may or may not be present in nudeoli which otherwise look identical and which, moreover, belong to nuclei showing similar distribution of chromatin material. They are not found in the less dense areas of nudeoli, and are quite unevenly distributed throughout the rest of the nudeolar material. They occur in pairs and small dusters of three or more. Whereas the above general description applies to nudeoli both from AlUum cepa and Vicia faba material, a number of significant differences have been observed in the distribution of the dense partides in these two plants, especially during certain stages of the mitotic cyde. During interphase (AlUum cepa), nudeoli usually show a few light spaces of various sizes within which no particles are found. There also often occurs a large central area which either appears empty or contains material resembling the chromatin masses in density and fine structural (Fig. 1). In such nudeoli, the particles are always localized in the outer dense material only. In Vicia faba two different types of interphase nudeoli have been found. The most common type

Structure and Mode of Formation of the Nucleolus in Meristematic Cells of Vicia faba and Allium cepa
Interphase nucleoli from Vicia faba and Allium cepa meristematic cells are roughly classified into two categories: (a) those that commonly show a rather homogeneous texture (except for small light spaces of various sizes) and frequently contain dense particles 140 A in diameter; (b) those found more frequently in Vicia characterized by a very sharp boundary between a dense outer cortex and a mu...
متن کاملA Light and Electron Microscope Study of Small, Spherical Nuclear Bodies in Meristematic Cells of Allium Cepa, Vicia Faba, and Raphanus Sativus
Interphase, preprophase, and prophase nuclei of meristematic cells of Allium cepa, Vicia faba, and Raphanus sativus are characterized by the presence of spherical bodies approximately 1 micro in diameter. These structures are Feulgen-negative but stain metachromatically with azure B, as the nucleolus, following fixation with glutaraldehyde. At the ultrastructural level, they consist predominant...
متن کاملSynthetic Capacities of Chromosome Fragments Correlated with Their Ability to Maintain Nucleolar Material
Onion (Allium cepa) and bean (Vicia faba) root tip cells containing many micronuclei, derived from x-ray-induced chromosome fragments, were exposed to H(3)-thymidine and H(3)-cytidine to determine the ability of such fragments to undergo DNA and RNA synthesis. Only a few micronuclei in onion and many in bean roots synthesize nucleic acid simultaneously with their main nuclei. A few micronuclei ...
متن کاملSensitivity of Allium cepa and Vicia faba towards cadmium toxicity
Sensitivity of common onion (Allium cepa L.) and faba bean (Vicia faba L.) to cadmium (Cd) stress was investigated using genotoxicity endpoints. Simultaneously, the antioxidative stress enzymes (guaiacol peroxidise and catalase) and lipid peroxidation [malonaldehyde (MDA) content] were investigated in the plants exposed to Cd in solution. The endpoints screened for genotoxicity included chromos...

Allium cepa, Guard Cell Protoplasts, Malate, Vicia faba Isolated and purified guard cell protoplasts of starch-containing ( Vicia faba) and starch-lacking (Allium cepa) stomata were used to study the metabolism of malate and Clanions as correlated with the K +-induced swelling of protoplasts. The correlation between swelling and malate synthesis in starch-containing and the inhibition of this c...
